Why is Cytokine Assay for Antibody Drugs Important?
Cytokines are small proteins produced by various cells in the immune system. They play a central role in regulating immune responses and inflammation. Cytokine release syndrome (CRS) is a rapid systemic inflammatory response characterized by secretion of pro-inflammatory cytokines by immune cells. Therefore, understanding the specifics of cytokine release is crucial for antibody drug development. Antibody drugs can inadvertently trigger a massive release of cytokines, a phenomenon known as a "cytokine storm", that can result in severe symptoms and sometimes fatal outcomes. Our Cytokine Assay Services
At our facility, the assay process begins with the isolation of primary human Peripheral Blood Mononuclear Cells (PBMC) or whole blood from healthy donors. In cytokine release assays, these cells are incubated with the antibody drug of interest. During incubation, the drug may bind to specific receptors on the immune cells, triggering these cells to release cytokines into the surrounding medium. After incubation, the cytokines in the medium are measured and quantified. Our assay utilizes the latest technologies and advanced platforms to provide accurate and reliable results.

Enzyme-Linked Immunosorbent Assay (ELISA) |
Enzyme-linked immunosorbent assays are the gold standard for cytokine detection and can quantify certain factors in the cytokine storm. In this method, the cytokines are detected by specific antibodies that bind to them. The bound antibodies are then detected by an enzyme-linked secondary antibody, which produces a color change when a substrate is added. The intensity of the color change is proportional to the amount of cytokine present. |
Multiplex Bead-Based Assays |
This method uses microscopic beads that are coated with specific antibodies. The beads are mixed with the sample, enabling the antibodies on the beads to capture the cytokines. Then, a secondary antibody is added, which is linked to a fluorescent dye. This dye emits a specific amount of light when it is agitated, and the amount of light is read by a special machine to quantify the amount of cytokine present. |
