PRM Quantitative Proteomics Services

Parallel Reaction Monitoring (PRM), an alternative method to SRM/MRM, is a targeted quantification method performed on hybrid quadrupole-Orbitrap (q-OT) and quadrupole time-of-flight (q-TOF) mass spectrometers. PRM has comparable sensitivity with similar linearity, precision, repeatability and dynamic range for protein quantification compared with SRM, but PRM provides high specificity, high selectivity, high accuracy and high-throughput quantification with confident targeted protein/peptide confirmation. It is ideal for quantifying tens to hundreds of targeted proteins/peptides in complex matrices.

PRM technology uses the quadrupole of Orbitrap or Q Exactive mass spectrometer to separate the target precursor ion before fragmenting the targeted precursor ion in the collision cell. The product ions can then be detected in the Orbitrap mass analyzer. This allows for accurate specificity analysis of the target proteins/peptides in complex samples.

Sample Requirement

Based on our special protein extraction technology, we can quickly extract proteins from various samples and formulate unique experimental schemes according to different research purposes. Specific requirements are as follows:

Sample TypeProteinCellAnimal TissuePlant TissueBloodUrineSerumMicrobes
Quantify100 ug1×107 cells1 g200 mg1 mL2 mL0.2-0.5 mLDry weighed: 200 mg

Note:

  • Please prepare enough dry ice or ice packs to ensure low temperature during sample delivery.
  • Our service is for research use only, and not for use in diagnostic procedures.
  • Plasma sample measurement requires at least 10 µl of plasma.

Key Features

  • Eliminates most interferences, providing more accuracy and attomole-level limits of detection and quantification
  • Broad dynamic range, covering 6 orders of magnitude of protein abundance at a time
  • Less assay development time
  • Absolute quantification of a variety of proteins without the need for antibodies
  • High-throughput, can identify up to 150 selected proteins at a time

Applications

  • Validation of protein abundance
  • Biomarker discovery and validation
  • Absolute quantitation, absolute quantitation of proteins, peptides, metabolites and serum, lipids from plasma and other biological samples
  • Protein post-translational modification analysis
  • Verification of untargeted proteomics (such as iTRAQ, label-free, SILAC) results
  • Pathological study of diseases

Reports

  • Experiment report
  • Relative parameters of the mass spectrometer
  • MS raw data files
  • Bioinformatics analysis
  • Peptide/Protein identification and intensity
