What is Protein Profiling Analysis

The Principle of Protein Profiling Analysis
There are many intact proteins of different molecular weights, solubilities and modifications in the samples. When using mass spectrometry to identify protein profiling, the proteins are digested into peptide fragments, which are then identified by shotgun proteomics. Shotgun proteomics is based on the principle that the proteins in the sample are first digested into peptides, which are then separated by HPLC. Next, the separated peptides are detected directly by primary mass spectrometry (MS1) and secondary mass spectrometry (MS2). MS1 measures the m/z and ion intensity of the peptide, and then the peptide is divided into different windows for MS2 detection. The peptide entering MS2 is fragmented, and then MS2 detects the ion fragments to obtain the peptide's spectral data. Compare the detected data with the appropriate databases, and the complete protein sequence can be obtained through analysis, thus realizing protein profiling analysis. Shotgun proteomics could acquire large amounts of data in a short period of time with its high-throughput assays, making it an excellent choice for protein profiling analysis.
