PTM Qualitative Analysis Service
Qualitative analysis of protein PTMs focuses on identifying the presence and nature of modifications rather than quantifying their abundance. It is crucial for understanding the diversity of PTMs, mapping modification sites, and gaining insights into cellular regulatory processes. Creative Proteomics offers high-quality PTM qualitative analysis services, including identification of modification types, sites and profiling, providing valuable insights into your protein research.
Methods for PTM Qualitative Analysis
PTM is the covalent process of altering proteins after protein biosynthesis. It can occur at the C- or N-terminus of amino acid side chains or proteins, and is performed by altering an existing functional group or by adding a new one to give peptide chains their diversity. PTM frequently occurs on functional groups that can act as nucleophiles in the reaction, such as: hydroxyl groups of serine, threonine, and tyrosine; amine groups of lysine, arginine, and histidine; sulfate anions of cysteine; carboxyl groups of aspartic acid and glutamic acid, etc.

Revealing the complex modifications of proteins and the dynamic essence of cellular processes, PTM qualitative analysis has become an essential research area in proteomics. Understanding PTMs is akin to deciphering a secret language written on proteins. Here are some commonly used methods for protein PTM qualitative analysis, such as:
Western Blotting
While commonly used for qualitative analysis, Western blotting provides information about the presence of specific PTMs. Antibodies targeting specific modified residues or motifs are used to detect modified proteins.
MS-based Techniques
MS is a powerful tool for qualitative PTM analysis. Tandem mass spectrometry (MS/MS) allows the fragmentation and sequencing of peptides, providing information about the presence and location of PTMs.
Antibody-based Techniques
Antibody microarray, IP and other antibody-based methods enable the qualitative measurement of specific PTMs. These assays provide high sensitivity and specificity for targeted modifications.
Applications of PTM Qualitative Analysis
- Biomarkers Discovery
Qualitative PTM analysis plays a crucial role in identifying protein modifications associated with diseases, such as the phosphorylation status of tumor markers. - Protein Structure and Function
Qualitative PTM analysis contributes to understanding how modifications influence protein structure and function, which contributes to revealing the relationship between protein modifications and their biological functions, providing a basis for drug design. - Signaling Pathways
Qualitative PTM analysis helps reveal important modification events in signaling pathways, such as phosphorylation of protein kinases or deacetylation events. - Drug Development and Treatment Strategies
Understanding the effects of drugs on protein modifications and how drugs regulate specific modifications aids in the development of more effective treatment strategies.
