What is UV-Vis used for?

UV-Vis spectroscopy is routinely used for the quantitative determination of different analytes, including biological macromolecules. Different molecules absorb ultraviolet or visible light of different wavelengths. An UV-Vis absorption spectrum will show a number of absorption bands corresponding to structural groups within the molecule.

How to perform UV-Vis analysis?

UV-Vis spectroscopy has been developed for quantitative analysis of biopharmaceuticals. In practice, a standard curve is constructed by plotting absorbance vs. molar concentration. Then the concentration of unknown compound is determined according to the measured absorbance. For example, protein can be measured at 280 nm directly, because of tryptophan and tyrosine. Two amino acid residues absorb UV strongly at 280 nm, and their content in many proteins are almost constant. Protein concentration also can be determined by other methods, such as Biuret method, Lower method, dye binding methods and turbimetric method.
