Fluorescent Neutralization Test

Antibody titers against a viral pathogen can be measured by neutralization assays. This assay is a specialized type of immunoassay and detects the presence of virus-neutralizing antibodies that are capable of inhibiting virus replication in a sample. To date, the neutralization assays include the traditional, such as the plaque reduction neutralization test (PRNT) and modern method, such as fluorescence reduction neutralization assay (FRNA). Virus neutralization test

For determination of antibody efficacy in many virus-caused diseases, virus neutralization tests are the gold standard. Among them, plaque reduction neutralization test (PRNT), which is based on plating virus-antibody mixture on virus-susceptible cells to detect and measure neutralizing antibodies, is the classical and most widely used method. However, the method is laborious and time-consuming due to its requirement of manual counting of plaques and immunostaining. Meanwhile, the operator bias and low throughput make it less suitable for large scale serodiagnosis and vaccine evaluation. Fluorescence reduction neutralization assay (FRNA) is an alternative to PRNT. Based on fluorescence measurement and analysis, the method can investigate neutralization of viruses by antibodies in an easier and faster way.
